Association of a Lipoxygenase Locus, Lpx-B1, with Variation in Lipoxygenase Activity in Durum Wheat Seeds

Alignment of a previously published cDNA lipoxygenase sequence (LoxA) from barley (van Mechelen et al., 1995) with four genomic lipoxygenase sequences amplified from the durum wheat varieties Jennah Khetifa and Cham 1. Boxed sequence represents coding sequence, and the sequence between the boxed regions is an intron. Shaded sequence corresponds to a Stowaway-type element discovered in the durum wheat sequences J4.2 and C7.1.2, but not in J2.2 and C5.36.2. Shaded sequence in italics is a direct repeat of the second half of the element, located immediately following the complete Stowaway element in C7.1.2 (see Fig. 2).

Fig. 2. Alignment of three Stowaway elements from previously published sequences in the Triticeae with the element sequences found in an intron of the lipoxygenase homolog in durum wheat. Black shading represents bases identical in at least half of the six compared sequences. On the first line is DMC1, an element from Australopyrum velutinum (Nees) B.K. Simon described as a Stowaway-type MITE element (Petersen and Seberg, 2000). On the second line is an element located in an intron of barley Knox3 gene (Mueller, et al., 1995). On the third line is an element located in an intron of barley BKIN12 protein kinase gene (Halford et al., 1992). Next, is the partial element sequence found in J4.2, a sequence from Jennah Khetifa. This is followed by the Stowaway element found in C7.1.2, which is from the Cham 1 variety of durum wheat. On the last line is the direct repeat of the second half of the element, located immediately following the complete Stowaway element in C7.1.2 (see Fig. 1).
