Growth of Red Clover Pollen. II. Elongation In Vitro1

Pollen of red clover (Trifolium pratense L.) frequently germinated up to 90% and formed pollen tubes with the modal length of 2.5 mm and the longest in the range from 5.0 to 10.0 mm while submerged in media which contained H3BO3. yeast extract, and Ca(NO3)2.4H2O at 50, 200, and 2,000 ppm, respectively, and sucrose at 35% w/v. The optimum sucrose concentrations for germination and elongation were 40 and 20%, respectively. The maximum germination and elongation occurred in the temperature range between 24 and 30 C. Germination and elongation were inhibited by agar (1%), KCN (10 ppm), and 6N-benzyladenine (20 ppm).
