The inheritance of resistance in Medicago sativa L. to the pea aphid, Macrosiphum pisi (Kalt.), was studied a resistant selection 81-1 and two susceptible selections. An antibiosis test for testing alfalfa plants for resistance to the pea aphid was developed using a controlled environment and a uniform stage of physiological growth. A quantitative score of the surviving aphids per adult day was a reliable index of plant resistance when considered in reference to a susceptible check variety or done. Progenies of the resistant selection 81-1 were classified either resistant or susceptible on the basis of surviving aphids per adult day. The results obtained from the S1 and S2 segregating progenies indicated that one dominant factor conferred resistance to the pea aphid. The S2 progeny reactions were in accordance with a tetrasomic interpretation. A hypothesis was presented concerning the behavior of the testcross progeny of S1 plants which permits agreement with this interpretation.
