Analysis of soybean seed lipoxygenases

Colorimetric assays for the three lipoxygenase isozymes in soybean [Glycine max (L.) Merr.] seed have been reported; however, they were not designed to analyze large numbers of samples. The objective of this study was to modify the assays to enable large-scale analysis. The information presented is based on the analysis of thousands of samples over several years for development of lipoxygenase-free soybean cultivars. The most significant change to the original procedure was the use of seed chips instead of homogenized soy flour, which has made possible large-scale analysis of seed samples with minimal resources. Other modifications to the original procedures and practical considerations with the use of the modified assays are discussed. Detailed descriptions of the assays are provided to facilitate their use by other researchers.
