A study was conducted to verify linkage between the loci Fr1 (root fluorescence) and Ep (seed coat peroxidase activity level) in the soybean, Glycine max (L.) Merr., as well as to test for linkage between the two loci and various other markers. Crosses were made between a standard cultivar (Minsoy) and a diverse selection of lines; the crosses included reciprocals and both linkage phases. The F1 plants were grown in five different location/year combinations. Segregation data for F2 confirm that Fr1 and Ep are linked. The combined estimate of linkage, based on 6734 F2 individuals, indicates that Fr1 and Ep are separated by a distance of 41.4 ± 0.8 cM. Genes Fr1 and Ep were tested for linkage with several cytological and genetic markers in segregating F2 populations. Linkage Group 12 is not physically linked to the trisomic chromosomes A, B, C, or D or to those chromosomes involved in a chromosome interchange from G. soja (Sieb. et Zucc.) PI 101404B. Linkage Group 12 also is independent of the loci Aco4, Dia1, Idh1, Pb, Sod1, Sp1, T, W1, Y9, and probably Ap.
