Assying for Linkage between Rps and Isozyme Loci in Soybean1

Linkage assays were run with soybean [Glycine max (L.) Merr.] lines differing for Rps and isozyme alleles, in an attempt to identify markers for Rps loci. The F2 seedlings were screened for resistance to specific races of Phytophthora megasperma (Drechs.) f.sp. glycinea (Hildeb.) Kuan & Erwin and cotyledon samples were assayed for isozyme patterns by means of starch gel electrophoresis. Alleles at the Rps1, Rps3, Rps4, Rps5, Rps6,, and Rps? loci were assayed for linkage with isozyme alleles at the Aco4, Dial, Idhl, IdH2, and Pgm1 loci. No linkages were detected for the 18 Rps-isoxyme pairs tested.
