Apparatus and techniques for the determination of photosynthetic carbon dioxide exchange and transpiration of individual leaves are described and evaluated. Stable, high rates of photosynthesis were obtained with attached leaves after preconditioning in the leaf chamber at high levels of illmnination. Particular attention was devoted to providing adequate air turbulence within the chamber and to operating variables, temperature and relative humidity, with regard to their overall effect on CO2 exchange in leaves.
